How do I plug in my guitar to my computer?

I've bought a good soundcard and physically I know how to plug the guitar in, the thing is I'm not sure of what to do with software, should I use cubase? or Reaktor? and even if I use one of them, what's next? how do I make the software realize that there's a guitar playing into the computer?
