" I knew it was you Fredo. You broke my heart. You broke my heart." from The Godfather2. It is the most dramatic scene I have ever seen. Al was great. And it is my favorite scene in the Godfather series.
Runner up is " Say hello to my little friend" from Scarface. Hey, another Al Pacino movie!

"You keep using that word, I do not think it means what you think it means"
-The Princess Bride
I use that quote a lot in my own life, that along with, "...I'm just not sure how well this plan was thought through..." from Meet the Robinsons, but that's another story.

I agree with the first person who answered. V for Vendetta has a great quote. Very memorable. But to be a little different I'm going to say a quote that barely any people will know, but I think was really powerful in it's movie.
It's in the movie Flicka. It's after the girls horse has been badly hurt by a cougar. The girl is sick in bed (she was with the horse and it was pouring rain). Her father is going to go shoot the horse, to put it out of it's misery.(on a side note he never really liked his daughter having the horse-it was wild_ The girl comes to the stairs as he's walking out with a rifle. He says something to the effect of, "I'm sorry." And the girl says, "It's okay daddy. You can shoot us."
When she said it my mouth dropped! I wasn't expecting us. But I think it is a very powerful, meaningful quote! It's great. *Of course the girl is literally expecting her dad to shoot her. She's saying that she and the horse are the same, they are one. In killing in the horse, he's killing her.
