Wait until the responses start to slow down (say, in a day or two). Then if no one else responds in that time, and you really like one of the answers you already got, then go ahead and give them Best Response. I haven't hit ten dollars yet, but this is my third(?) week here on myLot, and I've been taking it easy. I'm halfway there. I'm eager to see if I can get a payout in time for next month.

Well for the best response I used to wait one month but it is all up to you,if you read a response and you feel there will not be another one that can top it well then mark it. As far as hitting payout it all depends on how much you post,posting new discussions with good content is key also if you respond to to zero response discussions you earn more faster good luck and enjoy.
