How to make an effective application letter?

I just graduated from college and I've been eyeing this company I so want to get employed to. I'm now preparing the requirements to lodge an application, but I'm stuck with the application letter.
myLotter, any tips to help me craft an eye-catching application letter that would get me the job?

Sorry it is a bit late to come back to you but a good source of help would be your old college. They often have part time advisers in this art form even if not full time careers departments.
The only personal advice I have on the subject of CV's is keep them short and on a single page.
