In John's day (the writer of the book of rev/apocalypse.. he was referring to the tyranny of Rome.. as prophecy, today we have roman political architecture.. senates, courts, legislature.. all based on the Roman pattern.. And indeed the West was built on Roman/Greek foundations..

The number generally refers to something that is mentioned in the bible. It represents the "mark of the beast". Basically, the bible says it's a number of a man, 666. I've heard it explained as this: 6 is the number for man in a certain type of numerology. And, 3 is the number of God. So, 666 means that it is a man trying to be or claiming to be God. So, when people take this mark, it means they're pledging allegiance to this man. It's not a literal number, but a metaphor.

As far as I know, 666 is The Devil Number, like Omen had it on his head.
