Not really that major.
This season's finale (3x25, An Invisible Thread) Sylar will kill a major character, a character who's been around since season 1.
My guess it's Nathan, since Sylar has the shapeshifting ability. The Five Years Gone future shown in season 1 might be set in motion

I'm betting it's one of the Petrellis or Noah...although what a character says in the previews doesn't necessarily have anything to do with what is going to happen in the show, Sylar says Claire's going to be upset. But it's hard to guess which one because she's started seeing Angela as "Grandma." She always felt a strong connection to Peter but (since Milo and Hayden started dating, I noticed) they have kind of downplayed that a lot this season. No matter how many times he screws up, Noah is still Daddy but Nathan has started making an effort too.
So, if Sylar's comment has anything to do with the events of the next episode, it will probably be one of those four.
But if they continue to follow the "rules" of the comic book genre, Peter is still the closest thing Sylar has to a nemesis, even though his ability has changed, and a super-villain cannot exist without a direct opposite playing for the other side. So killing Peter without a replacement would make Sylar unstoppable which would kind of make fighting him pointless.

Since Peter's power is now a power that absorbs others powers temporarily he could easily be the only person to kill Sylar because all it take's is Peter and Sylar in the same room together and brush shoulders with Sylar then he would have Sylar's powers, but the thing is that Peter probably wouldn't know how to use half of the powers Sylar has now.So your right, Peter is still Sylar's nemesis and Sylar still isn't aware that Peter has any powers now, he still thinks Peter is without powers ( SURPRISE!..lol) But I wonder if they are going to do anything with Peter and Sylar before the end of this season.

I am not sure about that. The thing about heroes is that, they change totaly change what's going to happen. like Daphne. Matt was suppose to marry her in the future but it turns out that she will die. Well it seems that way. We will have to see the nex few episodes. The future where a lot of people would have abilities seems to be averted as well. The bomb in New York. Anyhting is possible. Nathan has been rumored to be remove in cast for quite sometime. Who knows. I guess will have to see how the story unfolds.
