As long as your mainboard has a PCI-E slot, then it supports PCI-E cards.
You'll have to check out the type of memory the graphics card uses, how much memory is has, and how much power it takes.
You'll have to get a graphics card that falls in your range. For example, you couldn't run a nVidia GTX 285 on a Dell Inspiron. Wrong type of memory, too much power, WAY too big, etc.
Also, if you have a slim case (I'm not that familiar with the gx270), you'll have to get a low profile card.
Also (LOL!), if your mainboard has onboard graphics, you'll have to go into the BIOS at startup and switch your settings so that your PC reads the PCI-E slot for your main graphics display, and also set your PC to actually turn the card on.
The GeForce GT 220 low profile is a great, affordable card. Radeon's X1300 low profile is also a good card.
The super high-end cards are really expensive, require a lot of memory, a lot of power and really aren't worth the money in my opinion.
I have a Inspiron 531s with 2GB RAM and a ATI Radeon X1300 and my graphics are awesome. I can play any game without a glitch.

The best way to know whether your PC can support a graphics card is by going to its manufacturer's website and doing the online search. Since you have Optiplex GX 270 by Dell, You can go to Dell's webpage. There is online detection facility. This software detects whether your PC is going to support a specific hardware or not. Remember, you can use the tag, which is called as service tag number, which can be found on a sticker on the front side of your PC.
