How to tell someone he is annoying in a nice way

I'm a career woman and sometimes, i really get annoyed with officemates who just can't seem to understand that there are more important tasks that needs to be done first. They are the types who follows you up every hour and get this, they can't take no for an answer. Am I being too nice or do I have to yell to make them get the point? Sometimes I just want to take the phone off the hook :-)

I think it is one of the things that most working people get disturbed by. I think that it should be good to be direct and tell them that you still have lots of things to do and just say sorry. They should understand, unless the one asking you to do something for him is your boss.
