could anyone explain what is the most important part or knowledge that need to practice or to learn in Microsoft excel?

It depends on what you're using it for. In business, the most important will probably be formatting, formulae (and therefore absolute addressing), names (cell referencing with sensible names), cross-tabs and pivot tables. Oh, and graphs. Everyone loves pretty pictures.

Formulae: stuff like "=A1+A2" or "=SUM(A1:A14)", that kind of thing.
