I'm a married man and also a manager at my place of work. Most of the people in my department are women. One of the responsibilities of the people who work for me is to fill out a time sheet telling how they spent their day and what projects they've worked on. Almost every day one of the ladies signs her time sheet XOXOXOXOXOXO (hugs and kisses) and she also writes things like, if you need me Friday night just give me a call, XOXOXOXO. Is she flirting with me and how can I tell her that I'm not interested without making a big deal out of this?

I think that you should have your wife talk to XOXOXOXOXOXOXO lady ... maybe she could meet her in the parking lot and let her know who she's dealing with! Though that could result in assault charges and you might have to bail your wife out of jail. I think there may be some validity to those who say that you need to protect yourself by allowing your boss/superiors know what's going on. I hate to say it, but since you're the man you'll probably have a harder time of proving that you're the one who's being harassed. If she does decide to spread rumors about you coming on to her, it's likely that people will believe the woman over you!

How do you tell her... easy.
Pull her a side, and tell her that this kind of behaviour is not accept at a place of employment. If it continues you will have to tell your boss. Also... I would recommend documenting all instances you have of the XOXOXOXOXO, and the date when you have the discussion say that you wish she would stop. If she does not stop, go to your boss with all the information and ask him to do something about it. If he does not, go to his boss.
Also tell her that you would also not appricate her action if it were out side of work.

I'm a married woman and I don't think that it's a compliment when other women want my husband. Actually I think it's offensive. I don't need another woman chasing after my husband to make me realize what a lucky woman I am. I am proud of my husband and the fact that I'm the only woman who could handle him!
