2 weks notice of resignation

can my employer just say your let go now, or do they have to respect the notice?

Normally, this is outlined in the employee handbook. Over all, if you ar ein the United States, most employers can let you go without notice. However, this will vary by state and quite often by company policy. Most employers have a set procedure in place that includes written and verbal warnings.
If you have resigned from your position, quite often an employer will allow you to stop working on the day you resign, but pay you for the next two weeks. If you work in the sales field or have constant contact with clients, this more likely than not will happen. They have to honor your two weeks notice and cannot force you to work past that two weeks. They can request that you stay until they find your replacement, but you are not required to do so.

What state do you live in? Michigan has "at will employment" no notice required to be fired or to quit.

Depending on what the reasons are for your leaving. (I'm assuming it's a resignation). Are you leaving for another job? Also, you may want to check your Company's Policies on resignations/separations, or whatever they may have discussed with you as part of the agreements when you were hired on.
Also consider the type of work you do. Does your position involve work activities that are unique? This may require special turnover of duties to the succeeding person.
