how to handle difficult people?

i am a people pleaser, not by choice, but i tend to be that way most of the time...and i really hate knowing i've hurt someone. however, there are times that i encounter people that are so difficult, literally...and i'm quite troubled if i could win them by staying quiet and handle their 'difficult' attitude in a calm way, or stand up to them. i will soon be having my first job, ever, and i'm hoping to make friends in my workplace without compromising my self-respect. have you had similar experiences? how have you handled them?
