The difference between DS and DSL is mostly the design, as melgashop stated. The DS is bulky, and the buttons are down low, putting more strain on your hands while you play. The mic is at the bottom forcing you to move your DS to use it which isn't helpful playing games like Yoshi Touch and Go, where you have to make good use of the mic. The Start and Select buttons are below the 4 normal buttons for easier access. And the largest difference is the stylus. DSL is on the side making it easy access for those fast pace games, or just easier to pull out while DS is opened.
Also being much larger, and heavier, making it more comfortable to use, especially for long periods of time.
DSL just looks better, a sleek smooth 'Mac' look and a plastic cover to give it that shiny look. Comes with a GBA slot place holder to allow it to keep the flush smooth design.
Other than the physical aspects of the two, the only difference is the brightness and colors. DSL is better than any handheld gaming system out to date, including the PSP. I'm not saying DSL is better than PSP, I don't want to start any thing, just saying the brightness is better.

Oh yikes. I can't say I'd pick either one. LOL. I'm still stuck on PS2 and Super Nintendo. The prices of the new fandangled ones is just too much for me!

Besides the design, the Nintendo DS Lite has a brighter screen than the old DS.
