I donate plasma on a regular basis. I don't know how other centers are run, but I go to the BioLife Plasma center. It's pretty simple in the long run and not very painful for me, but I have pretty big veins, which makes it easier for the people who have to stick you with the needle.
When you first go in, you normally have to take a physical(one physical anually at Biolife), you are then asked a series of medical questions first to see if you qualify that day for giving plasma. Then, on your first time, the check your veins to make sure that they're able to get the needle in there. Your temp, blood pressure and heart rate are taken. Then a sample of your blood is tested(normally through a small poke of the finger). The tests more or less check protien and iron levels in the blood.
Then your eventually taken to the donation area. The wrap your arm with a blood pressure monitoring strap. Then they swab the arm you use with iodine to disinfect the area After that, the strap inflates to cause veins to buldge. They then stick you with the needle. How much pain the needle acutally causes depends on how good your veins are of course, but once it's in, it's not really a problem.
More or less the machine goes through it's motions then. Seperate the plasma from the rest of the blood when it draws, and then injecting the seperated blood back into your systems along with a I.V. of something I can recall the name of at this time. You'll have to pump your fist/hand while the machine is taking and relax when it's giving it back. How much you have to give depends on your weight, but when you've filled your quota, they then take the plasma and your given a I.V. bag of saline to replace the plasma(feels cold going in, but not really all that bad).
The whole process, depending on the machine(some seem slower than others), how much you have to give based off of weight and how long you have to wait before donating if there's a line, can take between an hour to three.
That's pretty much how it's handled at the BioLife Plasma Center I go to.

I tried one time, but they said my veins were to small. I guess plasma is bigger than just regular blood (because I donate blood all the time) so they have to use a bigger needle when you donate plasma. Good luck with it though!!
