Other than the email notifications, we have the tabs above but you have to check them manually.
You can also add sorties to your homepage so that you can see who was last active on your topics.
You could also keep several tabs open to the discussions where you have participated and keep refreshing them. That is what I do and thankfully Chrome helps me a lot with this and rarely crashes

Above this discussion you will see 'discussions'.
Under that there are several tabs. Hover over it to see them and there you will see the discussions where you responded, started etc.
Your profile is one thing but I am referring to your home page: http://www.mylot.com/
There you can add sorties too.

There is a notification for friends who have been added to your list(requests) and you also get to receive a notification whenever you have a new message waiting for you. But.... there are no notifications for the responses we get from the comments or from the response we get from the discussions we created. I think if that happens that would have been a lot... =) imagine every actions you take in mylot will have an equal notification. =) But of course that would be great, at least we need not go to the discussions we can already see whatever there is that needs our actions. =)

There is only notifications for certain things, like messages, friends, etc. No notifications for people responding to your discussions (except in email account possibly?) but it certainly would be useful for carrying on conversations! Maybe someday they will implement it.
